About Helicase: The DNA Separator

- Helicase is an enzyme and motor protein that unwinds double-stranded deoxyribonucleic acid (DNA) into single strands.
- It is essential for DNA replication, repair, and transcription.
- Function of Helicase:
- DNA replication: It separates DNA strands so the genetic information can be copied.
- Access to genetic code: By unwinding DNA, it exposes the strands needed for cellular processes.
- Other roles: Helicase also plays an essential role in DNA repair and transcription.
- Working Mechanism of Helicase:
- ATP-powered movement: Helicase binds and hydrolyses ATP (Adenosine triphosphate), converting chemical energy into mechanical energy that helps it move along DNA.
- As helicase moves along the DNA molecule, it disrupts the hydrogen bonds between complementary bases, thereby separating the two DNA strands.
- This separation creates a Y-shaped replication fork, where the two DNA strands become exposed.
- As helicase moves along DNA, the DNA becomes increasingly twisted ahead of the replication fork.
- Other enzymes called topoisomerases relieve this twisting and prevent the DNA from becoming tangled or breaking.
- Once helicase separates the strands, replication machinery uses each exposed strand as a template to make a new complementary strand.
