News: Scientists in India have developed RK-251, a smart cancer drug that activates mainly inside cancer cells and may reduce harm to healthy cells.

  • RK-251 is a smart cancer drug designed to remain relatively inactive in normal tissues and activate in cancer cells.
  • Developed by: It was developed through collaborative research led by Dr. Asis Bala of the Institute of Advanced Study in Science and Technology (IASST) and Dr. K.P. Bhabak of the Indian Institute of Technology-Guwahati.
  • Working Mechanism:
    • Presence of ROS: Cancer cells often contain high levels of Reactive Oxygen Species (ROS), molecules that can cause cellular damage.
    • Activation of RK-251: When RK-251 enters a cancer cell, the high levels of ROS trigger its activation.
    • Release of anticancer compound: Once activated, RK-251 releases NBDHEX, an anticancer compound.
    • Action against cancer cells: NBDHEX targets proteins that many cancer cells use to survive and resist treatment, thereby producing its anticancer effect.
  • Key Features:
    • Targeted action: In preclinical studies, RK-251 showed strong activity against triple-negative breast cancer cells, with substantially lower effects on healthy cells.
    • Fluorescence response: The drug showed the desired fluorescence in the presence of ROS, indicating its activation in an ROS-rich environment.
    • Preclinical safety findings: Studies using zebrafish embryos (Danio rerio) showed no obvious signs of toxicity. 
    • Current Stage: RK-251 remains at the preclinical research stage, and further studies are required before human clinical trials.
